THE IMPORTANCE OF CLAUDINE 3, 4, 7 AT THE PROGNOSIS OF LARYNGEAL SQUAMOUS CELL CARCINOMAS

Objective: Claudines are tight junctions that provide connections between cells. In recent years a large number of studies have been conducted about the effect of the claudine- subtypes on the prognosis of cancer patients. Escepially, the prognostic value of claudine 7 on the oral squamous cell carcinomas has been revealed. As well as, there are also studies about the importance on colorectal, breast and prostate cancers. We aimed to investigate the staining patterns of histopathological samples with claudines of patients,whose postoperative pathology have been reported as laryngeal squamous cell carcinoma.

Material and method: 60 patients with laryngeal squamous cell carcinoma whose diagnosis and observation were made in Antalya Education and Research Hospital, Ear Nose Throat Departement,had been enrolled in the study.57 of the cases were men(95%) and 3 were women(5%).28 cases were in (46.6%)the early stage,and 32 cases were (53.3%) in the advanced stage.10% formaldehyde was used for tissue samples than these samples embedded in parafine and than histological sections were prepared.Light microscopic examinations were performed by applying hematoxylin eosin stain.The severity and extention of staining of claudine 3,4 and 7had been evaluated with immunohistochemical methods.

Results: The percentage of stainingof claudine7 in the advanced stage group was statistically significantly higher than the early stage group.The percentage of staining of claudine 7 in the early stage was 30% ; whereas the percentage of staining in the advanced stage was 55% . Independent from the other variables,each increase of 1% of the staining of the claudine 7,increases the possibility of being an advanced stage 1,021-fold.There was no significant difference in the percentage of staining in claudine 3 and 4.The intensity of staining of claudine 7 in the advanced stage group was statistically significantly higher than the early stage group.Independently from the other variables each increase of one-degree in the staining –intensity of claudine 7, increases the possibility of being an advanced stage 2.844-fold.The percentage of the intensity of staining of claudine 7 in the early stage was 78,6%;whereas the percentage of the intensity of staining of the advenced stage was 96,9%.There was no significant difference in the intensity of staining of claudine 3 and 4.

Conclusion: It has been determined, that the increase in the percentage of staining and in the intensity of staining of claudine 7 increases the possibility of being an advanced stage in patients with the diagnosis of laryngeal squamous cell carcinoma,. It was found, that the staining pattern of claudine 4 and 3, can not be used as a clinical and prognostic marker.
